Jour de canicule à New York. Au 51 Mulberry Street, la vie s'écoule normalement. Au même moment, plusieurs personnes, victimes de morsures de rats sont atteinte d'un mystérieux virus qu'elles transmettent à leur tour par morsure. Alors que la nuit tombe, toute la ville est contaminée, et New York est bientôt coupée du monde. Réfugiées dans leur immeuble de Mulberry Street, sept personnes tentent d'échapper à la contamination. A l'autre bout de la ville, une jeune femme doit rejoindre Mulberry Street en traversant Manhattan devenu un enfer de terreur et de mort. Tous vont vivre une terrible nuit...
